Application of Electrospun Polymer Membrane in Biomedical Devices

Abstract :Peritoneal adhesion is the most common adverse effect after abdominal surgery or inflammation. Till now, only solid barriers have been used successfully in clinical cases by acting as a physical separator during peritoneal healing. In this study, the casting film and fibrous membrane made from poly(lactic-co-glycolic acid) (PLGA) and polylactic acid-block-polyethylene glycol (PLA-b-PEG) mixtures were used and their chemical composition and performance have been studied in detail. They were observed to have almost identical adhesion-prevention performances as a commercial film consist of only PLGA, but have better flexibility and biocompatibility. Through the whole elaboration about this comprehensive and meticulous investigation from pathology, toxicity and immune response, the electrospun membrane with ECM-like (extracellular matrix) structure caused only normal acute inflammatory response and less intestinal obstruction. The feasibility, validity and safety were finally evaluated in a randomized, single-blind, placebo-controlled clinical trial. The statistical result of 150 patients confirmed that the fibrous membrane could reduce the incidence of abdominal adhesion between incision and organs directly underneath. A final approval from FDAC has been granted.
Several examples of further investigations on drug carrying and targeted delivery have been performed and will be discussed.
